SHORT BIO AND RESEARCH INTERESTS
I studied Biomedical Engineering (1999 - 2004) at the Politecnico di Milano (Milano, Italy), and graduated
in 2004 discussing a thesis entitled: “PhotoMEA: an opto-electronic biosensor for monitoring in vitro
neuronal network activity” (Supervisors: Prof. Giancarlo Ferrigno and Prof. Sara Mantero) that was awarded
as best master thesis from the National Group of Bioengineering. My master thesis work lead to the
development of a novel opto-electronic interface to investigate the activity of in-vitro neuronal networks
(EP Patent 1,842,049; US Patent 11/878,528; WO Patent 2,006,079,919). This neuro-artificial interface
combined a traditional multi-electrode array (MEA) with multisite optical stimulations/recordings of the
neuronal activity, thus improving the usage of chip arrays to investigate the properties of in-vitro cultured
neuronal networks (Ghezzi et al., BioSystems 2007).
At the beginning of 2005, I was appointed at the Bioengineering Department (Politecnico di Milano) with a
Ph.D. fellowship, granted from Fondazione Istituto Italiano di Tecnologia (IIT). I obtained my Ph.D. in
Bioengineering in 2008, discussing a thesis entitled: “Optical tools to stimulate in-vitro cultured
neurons” (Supervisors Prof. Giancarlo Ferrigno and Prof. Flavia Valtorta). During the Ph.D. course, I worked
as a guest student in Prof. Flavia Valtorta laboratory (San Raffaele Scientific Institute, DIBIT, Milano, Italy),
pursuing investigations on neuronal optical stimulation using caged-compounds, and on the development of

related interfaces (Ghezzi et al., Sensors 2008; Ghezzi et al., Journal of Neuroscience Methods 2008). Part
of my project was developed in collaboration with Ayanda Biosystems SA (EPFL, Lausanne, CH) where I
spent a useful research period. I also started a research project to develop a chamber for the long-term
stimulation and monitoring of electrical activity of in-vitro cultured neurons. The project was carried on by a
Ph.D. student, and was recently published (Biffi et al., Computational Intelligence and Neuroscience 2010;
Biffi et al., Biotechnology and Bioengineering 2012).
After the Ph.D., I joined the Neuroscience and Brain Technologies Department of IIT as Postdoctoral fellow
in the research group led by Prof. Fabio Benfenati. My main research line has been the development of an
organic retinal prosthesis to restore light sensitivity in cases of photoreceptor degeneration (Ghezzi et al.,
Nature Communications 2011; Ghezzi et al., Nature Photonics 2012). However, the multidisciplinary
scientific background that I developed during my scientific training also enabled me to participate in a
number of research collaborations:
1. The application of the high-density APS-MEA developed at IIT to acute brain slices for the spatial
analysis of plasticity events and epileptic paroxysms (Ferrea et al., Frontiers in Neural Circuits 2012),
for which I performed experiments comparing MEA recordings with voltage-sensitive dye imaging.
2. The evaluation of the effects of lithium as a cognitive enhancer in the Down syndrome (Contestabile et
al., The Journal of Clinical Investigation 2012), for which I performed the characterization of long-term
potentiation in the dentate gyrus of hippocampal slices.
3. The exploitation of optogenetic stimulation to investigate cross-modal interaction between primary
sensory areas (Iurilli G, Ghezzi D et al., Neuron 2012).
4. The development of a novel in-utero electroporation method for the specific expression of genes in
selected brain areas (Dal Maschio M*, Ghezzi D* et al., Nature Communications 2012, * first authors
with equal contribution).
My main expertise relates to optogenetics and optical stimulation combined to electrophysiology both invitro and in-vivo. My skills and competencies also include: (i) preparation of neuronal cell cultures, acute
brain slices and animals for in-vivo electrophysiology, (ii) in-vivo adult infection with lenti-viruses and
AAV-viruses in mice and rats, (iii) in-utero electroporation in rats, (iv) electrophysiological methods such as
micro-electrode arrays recordings, field recordings, tetrode arrays implant and patch-clamp recordings, (v)
voltage-sensitive, calcium-sensitive and 2-P imaging, (vi) fabrication of electronic and photonic devices,
(vii) design of electronic hardware for data management (such as amplifiers and DAQ systems), (viii)
development of software and firmware for system integration and signal processing, and (ix) managing of
various software for signal and imaging processing (e.g. MatLab, LabView and ImageJ)
I’m co-author of 12 peer-reviewed publications, 2 patents, 1 review, 1 book chapter, and 32 proceedings of
international congress. I supervised in the last 8 years: 9 master degree students, 2 PhD students and 1 Postdocs.
