Cap-Plus™ Detection Kit For Use with Capilliary Gap-Based Immunohistochemistry Staining Systems CAT. NO. 87-8143 FOR PRIMARY ANTIBODY Broad Spectrum* CHROMOGEN DAB GOOD FOR 600 slides *Will react with mouse, rabbit, guinea pig and rat primary antibodies. INTENDED USE For In Vitro Diagnostic Use. Interpretation must be made within the context of the patient’s clinical history and other diagnostic tests by a qualified pathologist. Invitrogen’s Cap-Plus™ Detection Kit is intended for use on automated capillary gap-based immunohistochemistry stainers such as the Ventana® TechMate®. This kit requires the use of the companion Cap-Plus™ Buffer Kit. Frozen or paraffin-embedded tissues, freshly prepared lymphocytes, and fixed cultured cells can be used. BACKGROUND The Cap-Plus™ Detection Kit uses Invitrogen’s 2nd Generation Labeled Streptavidin-Biotin (LAB-SA) detection technology. The LAB-SA method, also known as the Streptavidin-Peroxidase (SP) and Streptavidin-Alkaline Phosphatase (SAP) methods,(1) is illustrated in Figure 1. This method is currently preferred in the immunohistopathology laboratory due to its high sensitivity, low background, ease-of-use, and short protocol times. The interpretation of immunostaining results is complemented by positive and negative controls. Tissue specimens should be pre-incubated with blocking reagents to reduce nonspecific background immunostaining. Specific immunostaining is accomplished by localizing the antigens with a primary antibody. The antigen-antibody complex is identified using a LAB-SA biotinylated secondary antibody detection method. When using the LAB-SA method, a biotinylated secondary antibody will bind to the primary antibody that is complexed with the antigen. A streptavidin-enzyme conjugate is then added which binds to the biotinylated secondary antibody. A chromogen/ substrate solution is added that forms a colored deposit in the presence of the enzyme. The Cap-Plus™ Kit uses DAB substrate (3,3’ diaminobenzidine) as the chromogen. DAB results in the deposit of a brown insoluble precipitate at the antigenic sites containing the specific epitopes recognized by the primary antibody. The location of the antigen is then revealed by the presence of this colored deposit that forms around it. STORAGE AND SHELF LIFE Store kit at 2-8°C. All performance claims are void after kit expiration date. REAGENTS SUPPLIED Reagent A. One bottle (110 mL) of ready-to-use serum blocking solution Reagent B. One bottle (110 mL) of ready-to-use biotinylated secondary antibody Reagent C. One bottle (110 mL) of ready-to-use streptavidin-HRP-conjugated tertiary antibody Reagent D1. One dropper bottle (15 mL) of concentrated substrate buffer (20x) Reagent D2. One dropper bottle (15 mL) of concentrated chromogen solution, DAB (20x) Reagent D3. One dropper bottle (15 mL) of 0.6% hydrogen peroxide (20x) *Prepare DAB by adding 1 drop each of Reagents D1, D2, and D3 to 1 mL of H2O. Mix well. Protect from light and use within 4 hours. PRELIMINARY PREPARATION OF SLIDES -Appropriate tissue and antigen fixation is required to obtain reproducible performance and reliable 1. SPECIMEN PREPARATION 2. SLIDE PREPARATION interpretations. Suitable fixatives include: 10% neutral buffered formalin, B5, Bouin’s, Zinc formalin or alcohol-base fixatives. - Cell smears prepared from body fluids should be made to assure a monolayer of cells. Smears should be fixed immediately after preparation. Depending on the properties of the antigen, cell smears are usually stable for one to two weeks when stored at 4°C. - Fixation of cytospin or frozen sections can be done with acetone (100%) at 4°C for a period of 10 minutes. - To relax the specimens, place tissue sections into a container of deionized or distilled water preheated to 40-44oC. Then center the flat and wrinkle free sections onto a microscope slide treated with HistoGrip™. Proper placement will assure proper capillary action when two microscope slides are placed facing each other in the slide holder. -Precoat slides with HistoGrip™ (Invitrogen Cat. No. 00-8050). As an alternative, precoat with 0.1% polyL-lysine in water, then air dry. -Paraffin sections are deparaffinized with xylene and rehydrated in a graded series of ethanol. Wash cell smears or tissue in a PBS bath for 10 minutes before starting the staining procedure. 3. DEPARAFFINIZATION AND REHYDRATION Note: Do not allow specimen or tissue sections to dry from this point on. SUGGESTED STAINING PROTOCOL CONTINUED B. PROTOCOL FOR TECHMATE® AUTOMATED IMMUNOSTAINERS This kit is designed to be used in the same way as ChemMate® kits. In many cases, this kit can be used in your current protocol without any changes. The following steps may be used as a guideline for running the Cap-Plus™ Detection Kit on Ventana® TechMate® Automated Immunostainers. For complete details refer to the appropriate Ventana® TechMate® User Manuals. This Cap-Plus™ Detection Kit can also be used with other automated immunostainers using capillary gap technology. Please call Invitrogen’s Technical Service Department at 800-874-4494 for further details. PREPARATION FOR AUTOMATED IMMUNOSTAINING 1. Tissues should be mounted on microscope slides coated with HistoGrip™ (Invitrogen Cat. No. 00-8050) or poly-L-lysine. 2. Just prior to using the microscope slides, the tissue sections should be thoroughly dried with a conventional or microwave oven. This will eliminate the “water spotting” phenomenon when viewing the results. In a conventional oven, dry tissue sections on the microscope slides by heating them in an oven at 60oC for a minimum of 60 minutes. In an 800-watt microwave oven, place the microscope slides in a plastic slide holder at the center of the turntable and dry the slides for 2-3 on high power. Note: As a safety precaution place a separate water reservoir, at the same time, in the microwave oven. Carefully remove the slide holder and allow the melted paraffin to congeal before proceeding. 3. Deparaffinize tissue sections according to your approved laboratory procedures. (if not available see Table 2 in Appendix A). After deparaffinization, keep tissues wet in Buffer 2 from the Cap-Plus™ Buffer Kit. Slides should be immunostained on same day that they are deparaffinized. 4. Perform any required tissue preparation steps such as: epitope retrieval (see Table 3 in Appendix A), avidin/biotin blocking, tissue digestion, etc. Consult your antibody specification sheets. 5. Group microscope slides by antibody. 6. Determine the total number of slides to be run and then determine the total number of Ventana® TechMate® slide holders required. Use one slide holder per protocol. 7. Organize microscope slides for each Ventana® TechMate® slide holder using the Ventana® Antibody Positioning Sheet that is provided in the user’s manual. 8. Load slides according to Ventana® Antibody Positioning Sheet. 9. The microscope slides are now ready to begin automated immunostaining. CREATING THE USER DEFINED PROTOCOL 1. Status area of Map View Screen: move the mouse cursor to the column identified as LOAD. Click the mouse once. A predefined protocol list appears. 2. Find the MIP protocol by using the scroll bar. Move the cursor to this protocol. Click the mouse once to highlight. The MIP protocol should now appears in the File box. 3. On the smaller Load box, just below the File box, click once. A yellow blinking box appears on the Map View Screen indicating that this protocol is loaded. The mouse cursor can now be moved over to the position on the Map View Screen corresponding to the MIP protocol. Click the mouse once. 4. The desired template will immediately load and the Ventana® TechMate® tile area is labeled with the reagents that are required for each of the tiles. Also, the protocol is loaded. LOADING THE USER DEFINED REAGENT TRAYS 1. Complete an Antibody Positioning Sheet to determine which microscope slide goes into which slide holder. 2. Insert the microscope slide pairs, previously formed in the 10-well reagent trays, into the slide holder according to the Antibody Positioning Sheet. Note: The slide pairs will use the same primary antibody. 3. Prepare reagent trays and place them onto the “tiles” of the immunostainer’s platform. The number of trays prepared equals the number of paired slides to be run. The computer screen will show the template for reagent placement when the immunostainer’s protocol is loaded. Then, fill a 10-well tray with each of the following reagents according to Table 1. Table 1. Position the mouse pointer over the RUN button of the Status Area. Click the mouse once. The RUN button that turned red after the reagent protocol was loaded should now be green, indicating that the MIP immunostainer protocol has been started. If the slides were not counterstained on the TechMate®, after the TechMate® protocol has finished, the slides can be removed and manually counterstained and coverslipped if desired. (See Table 4 in Appendix A). SAFETY AND PRECAUTIONS 1. Take ample precautions when handling reagents. Use disposable gloves, coat, and safety glasses when handling suspected carcinogens. 2. After use, store as specified. Any storage conditions other than those specified in the package insert must be validated by the user. 3. Avoid contact of eyes and mucous membranes with these reagents. If these reagents come in contact with sensitive areas, wash with generous amounts of water. 4. DAB may be carcinogenic. Wear gloves when handling DAB and avoid contact with skin. Do not inhale or ingest. 5. Proclin (0.1%) is used as a preservative. Proclin is toxic if ingested. 6. Consult Federal, State or local regulations for disposal of any potentially toxic components. 7. Patient specimens and all materials coming into contact with them should be handled as if capable of transmitting infection, and disposed of with proper precautions. Never pipette by mouth, and avoid contact of this reagent and tissue specimens with skin and mucous membranes. 8. CLIA regulations should be followed in all applicable situations. 9. These reagents are optimally diluted. Los tejidos se deben montar en portas revestidos con HistoGrip™ (Invitrogen® Cat. Nº 00-8050) o poli-L-lisina. 2. Inmediatamente antes de utilizar los portas, se debe secar perfectamente los cortes de tejido con un horno convencional o un microondas. Esto eliminará el fenómeno de las "manchas de agua" cuando se visualicen los resultados. En un horno convencional, secar los cortes de tejido en el porta calentándolos a 60 oC por un mínimo de 60 minutos. En un horno a microondas de 800-W, colocar los portas en un portaobjeto plástico en el centro de la bandeja giratoria y secarlos durante entre 2 y 3 minutos a alta potencia. Nota: Como medida de seguridad, colocar en el horno a microondas, al mismo tiempo, un depósito separado con agua. Retirar el portaobjeto con todo cuidado y permitir que la parafina derretida se congele antes de proseguir. 3. Desparafinizar los cortes de tejido de acuerdo con los procedimientos aprobados por su laboratorio (si no se dispone de los mismos, consultar la Tabla 2 en el Apéndice B). Después de la desparafinación, mantener a los tejidos húmedos en Tampón 2 del Equipo tampón Cap-Plus™. Se debe realizar la inmunotinción de los portas el mismo día en que se los desparafina. 4. Seguir todos los pasos requeridos para la preparación del tejido: recuperación del epítopo (consulte la tabla 3 en el apéndice B), bloqueo de avidina/biotina, digestión tisular, etc. Consulte las hojas de especificación de los anticuerpos. 5. Agrupar los portas por anticuerpo. 6. Determinar el número total de portas que se van a analizar y luego establecer el número total de portaobjetos Ventana® TechMate® requeridos. Usar un portaobjeto por protocolo. 7. Organizar los portas para cada portaobjeto Ventana® TechMate® mediante la Hoja de posicionamiento de anticuerpos Ventana® que se proporciona en el manual del usuario. 8. Cargar los portas de acuerdo con la Hoja de posicionamiento de anticuerpos® Ventana. 9. Los portas ya están listos para comenzar la inmunotinción automatizada. Si los portas no fueron sometidos a doble tinción en el TechMate®, una vez concluido el protocolo TecMate® se pueden retirar y realizar la doble tinción de manera manual, colocando un cubreobjetos, si se desea. SEGURIDAD Y PRECAUCIONES 1. No escatime precauciones cuando maneje reactivos. Use guantes desechables, bata y gafas de seguridad cuando maneje materiales que se sospecha son carcinógenos. 2. Tras usar, almacene según se indica. Cualquier condición de almacenamiento distinta a la especificada en el paquete debe ser validada por el usuario. 3. Evite el contacto de ojos y mucosas con estos reactivos. Si estos reactivos entran en contacto con zonas sensibles, lave con abundante cantidad de agua. 4. DAB puede ser carcinogénico. Use guantes cuando maneje DAB y evite el contacto con la piel. No inhale ni ingiera. 5. Se usa Proclin (0,1%) como conservante. Proclin es tóxico por ingestión. 6. Consulte la normativa federal, estatal o local en materia de eliminación de compuestos potencialmente tóxicos. 7. Los especímenes de pacientes y todos los materiales que entren en contacto con ellos deberían manejarse como si fueran susceptibles de transmitir infecciones, y deberían eliminarse usando las debidas precauciones. No pipetee con la boca, y evite el contacto de este reactivo y de los especímenes tisulares con la piel y con las membranas mucosas. 8. Debería seguirse la normativa CLIA en todas las situaciones en las que sea de aplicación. 9. Estos reactivos se diluyen óptimamente. DÉPARAFFINAGE ET RÉHYDRATATION - Une fixation adéquate du tissu et de l’antigène est nécessaire pour obtenir une performance pouvant être reproduite et des interprétations fiables. Les fixateurs adéquats comprennent : formol tamponné neutre à 10 %, B5, formol de Bouin, de zinc ou des fixateurs à base d’alcool. - Des frottis de cellules préparés à partir de liquides organiques doivent être faits afin de garantir une couche unique de cellules. Ils doivent être fixés dès qu’ils sont prêts. Habituellement, en fonction des propriétés de l’antigène, les frottis de cellules restent stables pendant une à deux semaines lorsqu’ils sont conservés à 4 °C. - La fixation de sections congelées ou de cytospine peut être faite à l’acétone (100 %) à 4 °C pendant une période de 10 minutes. - Pour détendre les spécimens, placer les sections de tissu dans un récipient d’eau déionisée ou distillée préchauffée à 40-44 °C. Centrer ensuite les sections plates et lisses sur une lame de microscope traitée à l’HistoGrip™. Une bonne mise en place garantira une action capillaire adéquate lorsque deux lames de microscope seront placées l’une sur l’autre sur le porte-lames. - Enduire les lames de HistoGrip™ (Invitrogen No de cat. 00-8050). Une autre alternative consiste à les enduire de poly-L-lysine à 0,1 % dans de l’eau et de les sécher ensuite à l’air. - Les sections de paraffine sont déparaffinées avec du xylène et réhydratées dans une série d’éthanol classé. Laver les frottis de cellules ou le tissu dans un bain de soluté tampon de phosphate pendant 10 minutes avant de commencer la procédure de coloration. Ce kit de détection Cap-Plus™ peut également être utilisé avec d’autres immunocolorants automatisés utilisant la technologie de discontinuité capillaire. Les tissus doivent être attachés aux lames du microscope enduites de HistoGrip™ (Invitrogen No de cat. 00-8050) ou de poly-Llysine. 2. Les sections de tissu doivent être séchées à fond dans un four traditionnel ou à micro-ondes juste avant d’utiliser les lames du microscope. Ceci éliminera le phénomène de « taches d’eau » lors de l’examen des résultats. Dans un four traditionnel, sécher les sections de tissu sur les lames de microscope en les chauffant à 60 oC pendant 60 minutes minimum. Dans un four à micro-ondes de 800 watts, placer les lames du microscope dans un porte-lames en plastique au milieu du plateau rotatif et sécher les lames pendant 2 à 3 à puissance élevée. Remarque : par précaution, placer en même temps un récipient d’eau séparé dans le four. Retirer avec soin le porte-lames et laisser la paraffine fondue se solidifier avant de continuer. 3. Déparaffiner les sections de tissu en suivant les procédures approuvées par votre laboratoire (si elles ne sont pas disponibles, voir le tableau 2 à l’annexe B). Après le déparaffinage, garder les tissus humides dans le tampon 2 du kit de tampon Cap-Plus™. Les lames doivent être immunocolorées et déparaffinées le même jour. 4. Effectuer toute étape nécessaire de préparation du tissu telle que la récupération de l’épitope (voir le tableau 3 à l’annexe B), le blocage d’avidine/biotine, la digestion de tissu, etc. Se reporter aux fiches de spécification sur les anticorps. 5. Grouper les lames de microscope selon l’anticorps. 6. Déterminer le nombre total de lames à analyser et déterminer ensuite le nombre total de porte-lames Ventana® TechMate® nécessaires. Utiliser un porte-lames par protocole. 7. Organiser les lames de microscope pour chaque porte-lames Ventana® TechMate® en utilisant la feuille de positionnement des anticorps de Ventana® fournie avec le manuel de l’utilisateur. 8. Charger les lames conformément à la feuille de positionnement des anticorps de Ventana®. 9. Les lames de microscope sont désormais prêtes pour l’immunocoloration automatisée. CRÉATION DU PROTOCOLE DÉFINI PAR L’UTILISATEUR 1. Zone d’état de l’écran Indication cartographique : déplacer le curseur de la souris vers la colonne identifiée par LOAD (CHARGER). Cliquer une fois sur la souris. Une liste de protocoles prédéfinie s’affiche. 2. Localiser le protocole MIP en utilisant la barre de défilement. Déplacer le curseur vers ce protocole. Cliquer une fois sur la souris pour mettre en surbrillance. Le protocole MIP doit désormais apparaître dans la case File (Fichier). 3. Cliquer une fois sur la case plus petite Load (Charger) juste en dessous de la case File (Fichier). Une case jaune clignotante apparaît à l’écran Map View (Indication cartographique) indiquant que ce protocole a été téléchargé. Le curseur de la souris peut désormais être déplacé vers la position à l’écran Map View qui correspond au protocole MIP. Cliquer une fois sur la souris. 4. Le modèle désiré téléchargera immédiatement et la zone de mosaïque Ventana® TechMate® sera marquée avec les réactifs nécessaires pour chaque mosaïque. Le protocole est également téléchargé. Le bouton RUN (EXÉCUTER) doit être rouge afin d’indiquer que le protocole a bien été téléchargé. Compléter une feuille de positionnement des anticorps afin de déterminer quelle lame de microscope accompagne quel portelames. 2. Introduire les paires de lames de microscope, créées auparavant dans les plateaux de réactifs à 10 puits, dans le porte-lames, conformément à la feuille de positionnement des anticorps. Remarque : les paires de lames utiliseront les anticorps primaires. 3. Préparer les plateaux de réactif et les placer dans les « mosaïques » de la plate-forme de l’immunocolorant. Le nombre de plateaux préparés équivaut au nombre de paires de lames à analyser. L’écran de l’ordinateur affichera le modèle pour le placement du réactif une fois que le protocole de l’immunocolorant aura été chargé. Remplir ensuite un plateau à 10 puits avec chacun des réactifs suivants conformément au tableau 1. Tableau 1. La touche RUN (EXÉCUTER) qui avait viré au rouge lorsque le protocole de réactif a été chargé doit être vert, indiquant que le protocole de l’immunocolorant MIP a été lancé. Si les lames n’ont pas de coloration de contraste sur le TechMate®, une fois que le protocole TechMate® est terminé, les lames peuvent recevoir une coloration de contraste et être recouvertes à la main si on le souhaite. SÛRETÉ ET PRÉCAUTIONS 1. Faire très attention lors de la manipulation des réactifs. Porter des gants jetables, un tablier et des lunettes de protection lors de la manipulation de cancérogènes présumés. 2. Après l’utilisation, ranger tel que spécifié. Toute condition de stockage autre que celles spécifiées dans la notice de conditionnement doit être validée par l’utilisateur. 3. Éviter tout contact de ces réactifs avec les yeux et les muqueuses. Si ces réactifs entrent en contact avec des zones sensibles, laver abondamment avec l’eau. 4. La solution DAB peut être cancérogène. Porter des gants lors de la manipulation de la solution DAB et éviter tout contact avec la peau. Ne pas inhaler ou ingérer. 5. Le Proclin (0,1 %) est utilisé comme agent conservation. Le Proclin est toxique s’il est ingéré. 6. Consulter les règles fédérales, d’État ou locales pour l’élimination de tout composant potentiellement toxique. 7. Les échantillons et tous les matériaux avec lesquels ils entrent en contact doivent être manipulés comme étant susceptibles de transmettre une infection et doivent être éliminés en prenant les précautions nécessaires. Ne jamais remplir une pipette en aspirant par la bouche et éviter tout contact de ce réactif et des échantillons tissulaires avec la peau et les muqueuses. 8. Les règles CLIA doivent être suivies dans toutes les situations applicables. 9. Ces réactifs sont dilués de façon optimale. A seconda delle proprietà dell’antigene utilizzato, gli strisci di cellule rimangono solitamente stabili per una o due settimane se conservati a 4°C. - Il fissaggio di citospin o di sezioni congelate può essere eseguito con acetone (al 100%) a 4°C per un periodo di 10 minuti. - Per il rilassamento dei provini, immergere le sezioni tissutali in un contenitore di acqua deionizzata o distillata preriscaldata a 40-44oC. Quindi centrare le sezioni piatte e levigate su un vetrino da microscopio trattato con HistoGrip™. La centratura corretta garantisce un’azione capillare adeguata quando due vetrini da microscopio vengono inseriti nel portavetrini l’uno dirimpetto all’altro. - Pretrattare i vetrini con HistoGrip™ (N. Cat. Invitrogen 00-8050) Quale alternativa, pretrattare con poli-Llisina allo 0,1% in acqua, quindi lasciare asciugare spontaneamente. - Le sezioni incluse in paraffina devono essere deparaffinizzate con xilene e reidratate in una serie di alcoli selezionati. Lavare gli strisci di cellule o il tessuto in un bagno di PBS per 10 minuti prima di avviare la procedura di colorazione. In molti casi, questo kit può essere utilizzato nel proprio protocollo senza alcuna modifica. Quale linea guida per l’uso del Kit di rilevazione Cap-Plus™con gli immunocoloratori automatici Ventana® TechMate® si possono seguire le fasi riportate qui di seguito. Per informazioni particolareggiate ed esaustive pregasi consultare il Manuale per l’utente Ventana® TechMate®. Il presente Kit di rilevazione Cap-Plus™può essere usato anche con altri immunocoloratori automatici facenti ricorso alla tecnologia gap. Per ulteriori informazioni, pregasi rivolgersi al Reparto assistenza tecnica Invitrogen al numero 800-874-4494. PREPARAZIONE PER L’IMMUNOCOLORAZIONE AUTOMATICA 1. I tessuti dovrebbero essere fissati su vetrini da microscopio trattati con HistoGrip™ (N.®Cat. Invitrogen 00-8050) o poli-L-lisina. 2. Immediatamente prima dell’uso dei vetrini da microscopio, le sezioni tissutali dovrebbero essere completamente seccate in una stufa convenzionale o in un forno a microonde. Ciò consente di eliminare il fenomeno delle “macchiettature d’acqua” all’esame dei risultati. Se si usa una stufa convenzionale, seccare le sezioni tissutali sui vetrini da microscopio riscaldandole a 60oC per un minimo di 60 minuti. Se si usa un forno a microonde da 800-watt, inserire i vetrini da microscopio in un portavetrini di plastica e porli nel centro del piatto rotante, quindi farli asciugare per 2-3 minuti ad alta temperatura. Nota: quale misura precauzionale di sicurezza mettere allo stesso tempo nel forno microonde anche un serbatoio d’acqua. Estrarre con cura il portavetrini ed attendere che la paraffina sciolta si solidifichi prima di procedere. 3. Deparaffinizzare le sezioni tissutali conformemente alle procedure vigenti presso il proprio laboratorio (qualora non siano disponibili, pregasi consultare la Tabella 2 nell’Appendice B). Eseguita la deparaffinizzazione, lasciare i tessuti immersi nel Tampone 2 del Kit per tampone Cap-Plus™. L’immunocolorazione dei vetrini dovrebbe essere eseguita lo stesso giorno in cui vengono deparaffinizzati. 4. Eseguire tutte le fasi di preparazione del tessuto necessarie, quali: richiamo dei determinanti antigeni (vedasi la Tabella 3 nell’Appendice B), blocco dell’avidina/biotina, digestione del tessuto, ecc. Pregasi consultare la scheda tecnica relativa all’anticorpo usato. 5. Raggruppare i vetrini da microscopio per anticorpo. 6. Stabilire il numero complessivo di vetrini che si intende utilizzare e quindi determinare il numero complessivo di portavetrini Ventana® TechMate® occorrenti. Usare un portavetrini per protocollo. 7. Organizzare i vetrini da microscopio per ciascun portavetrini Ventana® TechMate®facendo riferimento alla Scheda di posizionamento degli anticorpi Ventana®riportata nel manuale per l’utente. 8. Caricare i vetrini conformemente a quanto riportato nella Scheda di posizionamento degli anticorpi Ventana®. 9. I vetrini da microscopio sono ora pronti e si può quindi avviare l’immunocolorazione automatica. MESSA A PUNTO DEL PROTOCOLLO DEFINITO DALL’UTENTE 1. Area status dello Schermo Map View (Visualizzazione mappa): spostare il cursore del mouse sulla colonna intitolata LOAD (Carica). Fare clic con il mouse una volta. Compare un elenco predefinito di protocolli. 2. Individuare il protocollo MIP usando la barra di scorrimento. Spostare il cursore su questo protocollo. Fare clic con il mouse una volta per evidenziarlo. Il protocollo MIP dovrebbe ora apparire nella casella File. 3. Fare clic una volta sulla casella Load (Carica) più piccola, appena al di sotto della casella File. Nello schermo Map View (Visualizzazione mappa) compare una casella gialla lampeggiante ad indicazione dell’avvenuto caricamento del protocollo selezionato. Si può ora spostare il cursore del mouse sulla posizione all’interno dello schermo Map View (Visualizzazione mappa) corrispondente al protocollo MIP. Fare clic con il mouse una volta. 4. Il modello desiderato viene caricato immediatamente e l’area dei tile Ventana® TechMate® appare etichettata con i reagenti richiesti per ciascun tile. Viene inoltre caricato il protocollo. Il tasto RUN (Esegui) dovrebbe ora essere rosso ad indicazione dell’avvenuto caricamento del protocollo. Inserire le coppie di vetrini da microscopio, precedentemente create in vaschette per reagenti da 10 pozzetti, nel portavetrini conformemente a quanto riportato nella Scheda di posizionamento degli anticorpi. Nota: le coppie di vetrini usano lo stesso anticorpo primario. 3. Preparare le vaschette dei reagenti ed appoggiarle sui “tile” della piattaforma dell’immunocoloratore. Il numero di vaschette preparate coincide con il numero di vetrini accoppiati che si intende utilizzare. Lo schermo del computer mostrerà il modello per il posizionamento dei reagenti al caricamento del protocollo dell’immunocoloratore. Quindi, riempire una vaschetta da 10 pozzetti con ciascuno dei seguenti reagenti, conformemente a quanto riportato nella Tabella 1. Tabella 1. Se i vetrini non sono stati controcolorati sul TechMate®, al termine del protocollo TechMate®, possono essere rimossi e controcolorati manualmente e, se lo si desidera, inseriti in un coprivetrino. SICUREZZA E PRECAUZIONI 1. Esercitare molta cautela durante la manipolazione dei reagenti. Indossare guanti monouso, un camice e occhiali protettivi durante la manipolazione di sostanze di cui si sospetta la cancerogenicità. 2. Dopo l’uso, conservare conformemente a quanto specificato. Qualsiasi altra condizione di conservazione che si discosti da quelle specificate nel foglietto illustrativo deve essere convalidata dall’utente. 3. Evitare il contatto dei reagenti con gli occhi o con le mucose. In caso di contatto accidentale dei reagenti con aree sensibili, risciacquare con acqua abbondante. 4. La DAB potrebbe essere una sostanza carcinogenica. Indossare dei guanti quando si manipola la DAB ed evitare il contatto con gli occhi. Non inalare o ingerire. 5. Proclin (allo 0,1%) è usato quale conservante. Proclin è tossico se ingerito. 6. Fare riferimento alle normative vigenti a livello nazionale o locale in materia di smaltimento di sostanze potenzialmente tossiche. 7. I provini dei pazienti e tutti i materiali che entrano in contatto con essi devono essere manipolati quali materiali potenzialmente infettivi e quindi smaltiti adottando le debite precauzioni. Non pipettare mai con la bocca ed evitare il contatto di questo reagente e dei provini tissutali con la cute e le mucose. 8. Si devono osservare i regolamenti previsti dalla legge statunitense per il miglioramento dei laboratori clinici (Clinical Laboratory Improvement Act, CLIA) in tutte le situazioni applicabili. 9. I presenti reagenti sono stati diluiti in base al rapporto ottimale. VORLÄUFIGE VORBEREITUNG DER OBJEKTTRÄGER -Um reproduzierbare Ergebnisse und zuverlässige Interpretationen zu erhalten, ist eine angemessene Gewebe1. PROBENVORBEREITUNG 2. VORBEREITUNG DER OBJEKTTRÄGER 3. ENTPARAFFINISIERUNG UND REHYDRIERUNG und Antigen-Fixierung erforderlich. Geeignete Fixierungsmittel sind u.a.: 10% neutrales gepuffertes Formalin, B5, Bouin’s, Zinkformalin oder Fixierungsmittel auf Alkoholbasis. - Es sollten Zellabstriche von Körperflüssigkeiten gemacht werden, um eine Monozellschicht zu erhalten. Die Abstriche sollten unmittelbar nach der Präparation fixiert werden. Je nach den Eigenschaften des Antigens sind die Zellabstriche gewöhnlich für ein bis zwei Wochen stabil, wenn sie bei 4°C aufbewahrt werden. - Die Fixierung von Cytospin oder gefrorenen Gewebeschnitten kann mit Azeton (100%) bei 4°C für eine Dauer von 10 Minuten erfolgen. - Um die Proben zu entspannen, die Gewebeschnitte in einen Behälter mit deionisiertem oder destilliertem Wasser geben, das auf 40-44°C erwärmt wurde. Dann die flachen und faltenlosen Gewebeschnitte auf einen mit HistoGrip™ behandelten Objektträger aufziehen. Die sachgemäße Platzierung gewährleistet eine angemessene Kapillarwirkung, wenn zwei Mikroskop-Objektträger auf dem Objektträgerhalter einander gegenüber platziert werden. -Die Objektträger mit HistoGrip™ (Invitrogen Kat.-. Nr. 00-8050) beschichten. Als Alternative können sie auch mit 0,1% Poly-L-Lysin in Wasser beschichtet und dann an der Luft getrocknet werden. -Die Paraffinschnitte werden mit Xylen entparaffinisiert und in einer gradierten Serie von Ethanol rehydriert. Vor dem Beginn des Färbungsverfahrens die Zellabstriche oder Gewebe für 10 Minuten in einem PBS-Bad waschen. PROTOKOLL FÜR AUTOMATISIERTE TECHMATE® IMMUNOSTAINER Dieses Kit wird genau so verwendet wie die ChemMate® Kits. In vielen Fällen kann dieses Kit ohne Änderungen im aktuellen Protokoll verwendet werden. Die folgenden Schritte dienen als Richtlinie für den Gebrauch der Cap-Plus™ Detektionskits auf automatisierten Ventana® TechMate® Immunostainern. Vollständige Details sind in der entsprechenden Gebrauchsanleitung des Ventana® TechMate® zu finden. Dieses Cap-Plus™ Detektionskit kann auch mit anderen automatisierten Immunostainern, die Kapillarspalttechnologie anwenden, verwendet werden. Weitere Details sind von Invitrogens technischem Kundendienst unter +1-800-874-4494 (gebührenfrei innerhalb der USA) zu erfahren. VORBEREITUNG FÜR AUTOMATISIERTES IMMUNOSTAINING 1. Die Gewebe sollten auf mit HistoGrip™ (Invitrogen Kat.- Nr. 00-8050) oder Poly-L-Lysin beschichtete Mikroskop-Objektträger aufgezogen werden. 2. Unmittelbar vor dem Gebrauch der Mikroskop-Objektträger sollten die Gewebeschnitte in einem normalen Ofen oder einem Mikrowellenherd gründlich getrocknet werden. Dadurch wird bei der Untersuchung der Ergebnisse das "Wasserfleck"-Phänomen vermieden. In einem normalen Ofen die Gewebeschnitte auf den Mikroskop-Objektträgern bei 60°C für eine Dauer von mindestens 60 Minuten trocknen lassen. In einem 800-Watt-Mikrowellenherd die Mikroskop-Objektträger in einen Plastik-Objektträgerhalter geben, der auf die Mitte der Drehscheibe gestellt wird, und bei höchster Stufe 2-3 Minuten lang trocknen lassen. Hinweis: Als Sicherheitsmaßnahme gleichzeitig einen separaten Wasserbehälter in die Mikrowelle stellen. Den Objektträgerhalter vorsichtig herausnehmen und vor dem Fortfahren das geschmolzene Paraffin fest werden lassen. 3. Die Gewebeschnitte gemäß den genehmigten Laborverfahren entparaffinisieren (wenn keine vorhanden sind, siehe Tabelle 2 in Anhang B). Nach der Entparaffinisierung die Gewebe in Puffer 2 des Cap-Plus™ Pufferkits feucht halten. Die Objektträger sollten noch am gleichen Tag, an dem Sie entparaffinisiert wurden, immungefärbt werden. 4. Alle erforderlichen Gewebevorbereitungsschritte durchführen wie: Epitop-Retrieval (siehe Tabelle 3 in Anhang B), Avidin/BiotinBlocking, Gewebe-Digestion, usw. Konsultieren Sie das Antikörper-Datenblatt. 5. Die Mikroskop-Objektträger nach Antikörper gruppieren. 6. Bestimmen, wieviele Objektträger insgesamt verarbeitet werden sollen und dann die insgesamt erforderliche Anzahl an Ventana® TechMate® Objektträgerhaltern bestimmen. Pro Protokoll einen Objektträgerhalter verwenden. 7. Mit Hilfe des in der Gebrauchsanleitung enthaltenen Ventana Antikörper-Positionierblatts die Mikroskop-Objektträger für jeden Ventana® TechMate®-Objektträgerhalter organisieren®. 8. Die Objektträger dem Ventana® Antikörper-Positionierblatt gemäß laden. 9. Die Mikroskop-Objektträger sind jetzt bereit, das automatisierte Immunostaining zu beginnen. ERSTELLEN DES VOM BENUTZER DEFINIERTEN PROTOKOLS 1. Statusbereich des Map View-Bildschirms: den Mauscursor zu der als LOAD identifizierten Spalte bewegen. Die Maus einmal klicken. Ein vordefiniertes Protokoll erscheint. 2. Mit der Bildlaufleiste das MIP-Protokoll suchen. Den Cursor zu diesem Protokoll bewegen. Die Maus einmal klicken, um das Protokoll zu markieren. Jetzt sollte das MIP-Protokoll im File-Feld erscheinen. 3. Einmal auf das kleinere Load-Feld unterhalb des File-Felds klicken. Auf dem Map View-Bildschirm erscheint ein gelbes blinkendes Feld, das anzeigt, dass dieses Protokoll geladen ist. Der Mauscursor kann jetzt zu der Position auf dem Map View-Bildschirm bewegt werden, die dem MIP-Protokoll entspricht. Die Maus einmal klicken. 4. Die gewünschte Schablone wird automatisch geladen und der Ventana® TechMate® Kachelbereich ist jetzt mit den Reagenzien markiert, die für jede Kachel erforderlich sind. Das Protokoll ist auch geladen. Die RUN-Schaltfläche sollte jetzt rot sein, um ein erfolgreiches Laden des Protokolls anzuzeigen. Ein Antikörper-Positonierblatt ausfüllen, um zu bestimmen, welcher Mikroskop-Objektträger in welchen Objektträgerhalter gehört. 2. Die zuvor in den 10-Mulden-Reagenztabletts geformten Mikroskop-Objektträgerpaare dem Antikörper-Positionierblatt gemäß in den Objektträgerhalter einsetzen. Hinweis: Die Objektträgerpaare müssen die gleichen Antikörper verwenden. 3. Die Reagenztabletts vorbereiten und sie auf die "Kacheln" der Immunostainer-Plattform platzieren. Die Anzahl der vorbereiteten Tabletts entspricht der Anzahl der zu verarbeitenden Objektträgerpaare. Auf dem Computerbildschirm erscheint die Schablone für die Reagenzplatzierung, wenn das Protokoll des Immunostainers geladen ist. Dann ein 10-Mulden-Tablett gemäß Tabelle 1 mit jedem der folgenden Reagenzien füllen. Tabelle 1. Die Maus einmal klicken. Die RUN-Schaltfläche, die nach dem Laden des Reagenzprotokolls rot wurde, sollte jetzt grün sein und anzeigen, dass das MIP-Immunostainer-Protokoll gestartet wurde. Wenn die Objektträger nicht auf dem TechMate® gegengefärbt wurden, nachdem das TechMate®-Protokoll beendet wurde, können die Objektträger falls erwünscht entfernt und manuell gegengefärbt und mit einem Deckglas bedeckt werden. SICHERHEIT UND VORSICHTSMASSNAHMEN 10. Bei der Handhabung der Reagenzien ausreichende Sicherheitsmaßnahmen ergreifen. Beim Umgang mit angenommenen Karzinogenen Einweghandschuhe, Laborkittel und Augenschutz tragen. 11. Nach dem Gebrauch wie vorgeschrieben aufbewahren. Alle Aufbewahrungsbedingungen, ausser den in der Packungsbeilage vorgeschriebenen Bedingungen, müssen vom Benutzer validiert werden. 12. Diese Reagenzien nicht mit den Augen oder Schleimhäuten in Kontakt bringen. Bei einem Kontakt dieser Reagenzien mit empfindlichen Bereichen diese mit reichlich Wasser abwaschen. 13. DAB kann karzinogen sein. Bei der Handhabung von DAB Handschuhe tragen und einen Hautkontakt vermeiden. Nicht einatmen oder einnehmen. 14. Proclin (0,1%) dient als Konservierungsmittel. Proclin ist bei Einnahme toxisch. 15. Bei der Entsorgung potenziell toxischer Bestandteile die staatlichen bzw. örtlichen Bestimmungen beachten. 16. Proben von Patienten und alle Materialien, die mit diesen in Kontakt kommen, sollten als möglicherweise infektiös gehandhabt und mit angebrachten Sicherheitsvorkehrungen entsorgt werden. Niemals mit dem Mund pipettieren und einen Kontakt dieses Reagenz und der Gewebeproben mit der Haut und den Schleimhäuten vermeiden. 17. In allen Situationen sollten die Vorschriften der CLIA eingehalten werden. 18. Diese Reagenzien sind optimal verdünnt.
