Come effettuare una raccolta
del materiale seminale
How to collect the equine semen

Anche se la raccolta del seme da uno stallone è una procedura relativamente facile, la
qualità dell’eiaculato può venire relativamente influenzata dalle procedure utilizzate per
l’operazione. Il tipo di vagina artificiale scelta per il cavallo sarà determinato dalla preferenza dello stallone oltre che da quella del veterinario. Alcuni stalloni sono abituati ad una
vagina artificiale dalle pareti morbide, mentre altri si trovano maggiormente a loro agio
con una rigida, come la Colorado model AV.
La vagina artificiale deve essere regolata in
modo da adattarsi a stalloni di varie taglie e
deve essere costruita in modo da suscitare
una risposta favorevole. Lo stallone eiacula
in base a temperatura e pressione. Quindi,
questi due parametri devono essere regolati
individualmente per ciascun maschio. La
temperatura della vagina artificiale prima
della raccolta deve essere compresa fra 42 e
48 °C. Lo strumento deve essere montato con
un anticipo sufficiente a consentire alla temperatura di stabilizzarsi prima della raccolta.
Per la preparazione della vagina artificiale si
utilizza una piccola quantità di lubrificante.
Recenti studi hanno dimostrato che grandi
quantità di lubrificanti idrosolubili possono
influire sulla qualità del seme. La bottiglia
ideale per la raccolta è quella in cui è possibile collocare un filtro in linea. Ciò consente
di separare il seme nella frazione ricca di
spermatozoi e in quella gel. Quest’ultimo
viene solitamente scartato e per la valutazione e l’inseminazione si utilizza soltanto la
frazione ricca di spermatozoi.

Although semen collection from a stallion
is a relatively easy procedure, the quality of
the ejaculate can be affected dramatically by
the procedures used for the collection of semen. The type of artificial vagina selected for
the horse will be determined by the stallion’s
preference, as well as the preference of the
veterinarian. Some stallions are accustomed
to a soft-sided artificial vagina, whereas others are more comfortable using a hard-side
artificial vagina, such as the Colorado model
AV. The artificial vagina should be adjusted to
accommodate stallions of various sizes and
constructed so to elicit a favorable response.
The stallion ejaculates based on temperature and pressure. Thus, these two parameters
must be adjusted individually for each stallion. The temperature of the artificial vagina
prior to collection should be in the range of
42 to 48°C. The artificial vagina should be
constructed well enough in advance so that
the temperature has stabilized prior to collection. A small amount of lubricant should be
used for preparing the artificial vagina. Recent studies have shown that large quantities
of water-soluble lubricants can affect the
quality of the semen. The ideal collection bottle is one in which an inline filter can be positioned. This allows the semen to be separated into the sperm-rich fraction and the gel
fraction. The gel usually is disposed of and
only the sperm-rich fraction is used for evaluation and insemination.
The collection shed should be of adequate
size to allow the stallion to have room to maneuver, as well as to allow one to position a
jump mare within the collection shed. Semen

permettere di introdurvi un manichino di fattrice. La raccolta del seme può essere effettuata mentre lo stallone monta un manichino
o una fattrice viva. Indipendentemente dal
metodo utilizzato, per lo stallone è estremamente importante l’equilibrio. Inoltre, il pavimento del locale di raccolta deve essere
realizzato con un materiale che possa essere
disinfettato e pulito a fondo. Generalmente,
risulta ideale sia per garantire l’equilibrio che
per la pulizia un pavimento coperto da tappetini o altro materiale in gomma o, eventualmente, da gomma sminuzzata.
Una volta che la vagina artificiale sia alla
temperatura giusta, si posiziona dietro una
barriera una fattrice destinata a stimolare il
maschio e si introduce lo stallone nel locale
destinato al prelievo. Dopo aver presentato il
cavallo alla fattrice in estro e aver ottenuto
l’erezione, si lavano con acqua calda il fusto
del pene ed il glande. Se lo stallone è destinato a montare un manichino, una volta ottenuta un’erezione, dopo il lavaggio viene avvicinato alla falsa fattrice. È auspicabile che
lo stallone sia stato abituato a montare il manichino senza che accanto ad esso sia stata
posta una cavalla viva. Una volta sul manichino, si devia il pene dello stallone introducendolo nella vagina artificiale. Quest’ultima
viene tenuta in posizione tale da risultare diretta leggermente verso l’alto, in modo da simulare l’angolazione dell’apparato riproduttore della fattrice. Se lo stallone non è abituato al manichino, il prelievo va effettuato
servendosi di una fattrice viva in estro. Questa può essere posizionata al centro del locale ed esposta allo stallone. In condizioni
ideali il maschio dovrebbe manifestare un’erezione e montare la fattrice da dietro. Tuttavia, alcuni stalloni sono abituati a porsi accanto alla fattrice per poi retrocedere e montarla. Una volta che lo stallone sia montato
sulla fattrice, si devia lateralmente il pene e
lo si introduce nella vagina artificiale. È possibile posizionare la parte piana della mano
sull’uretra per percepire i getti di sperma che
la percorrono. Una volta che attraverso l’uretra siano passati tre o quattro getti di sperma
la vagina artificiale viene lentamente abbassata in modo che si venga a trovare quasi ver-

to the estrous mare and, upon obtaining an
erection, the shaft of his penis and glans are
washed with warm water. If the stallion is to
be mounted on a phantom, once he obtains an
erection, after washing he is presented to the
phantom. Hopefully, the stallion has been
trained to mount the phantom without having
a mare positioned alongside the phantom.
Once upon the phantom, the stallion’s penis is
deflected and placed into the artificial vagina.
The artificial vagina is held in a slightly upward position to simulate the angle of the
mare’s reproductive tract. If the stallion is not
trained to the phantom, then he must be collected off of a live estrous mare. The mare can
be positioned in the middle of the breeding
shed and the stallion presented to the mare.
Ideally, the stallion would obtain an erection
and mount the mare from the rear. However,
some stallions are accustomed to coming
alongside the mare and then backing up and
mounting the mare. Once the stallion is
mounted on the mare, the penis is deflected to
the side and introduced into the artificial vagina. One can position the flat part of the hand
onto the urethra and feel the jets of semen
moving through the urethra. Once three or
four jets of semen have passed through the
urethra, the artificial vagina is slowly lowered
so that it is nearly vertical by the end of the
ejaculatory process. This allows the semen to
drain through the filter in the collection bottle
to the bottom of the collection bottle.
It is very important to empty approximately two-thirds out of the artificial vagina immediately after ejaculation. This releases the
pressure and accommodates the semen drain-

del materiale ottenuto può venire prelevata
dalla bottiglia di raccolta e destinata all’analisi per valutare mobilità, numero e concentrazione degli spermatozoi.
