Invitrogen PCNA STAINING KIT CAT. NO. 93-1143 GOOD FOR 50 slides INTENDED USE For In Vitro Diagnostic Use. Interpretation must be made within the context of the patient’s clinical history and other diagnostic tests by a qualified pathologist. PCNA Staining kits are designed to reveal antigen on frozen or paraffin-embedded tissues, freshly prepared lymphocytes, and fixed cultured cells. BACKGROUND PCNA (Proliferating Cell Nuclear Antigen) was previously known as cyclin. PCNA is a 36 kDa nonhistone protein found in the nucleus that plays a role in the initiation of cell proliferation by mediating DNA polymerase. PCNA levels are elevated in the S, G2, and M phases of cell mitosis in normal and malignant tissues. PCNA expression has a broad correlation with mitotic activity and can be used as a marker for cell proliferation. PCNA has proven useful for proliferative studies of normal and neoplastic tissues both in vivo and in vitro (1-7). Invitrogen’s PCNA staining kit uses a biotinylated PCNA monoclonal antibody (clone PC10), thus eliminating the need for a species-specific secondary antibody. As a result, PCNA staining is exceptionally clean and can be performed in tissue and cell samples from most species, including: human, primate, mouse, rabbit, rat, yeast, and insect without background caused by cross-reactivity with endogeneous immunoglobulins. Streptavidin-peroxidase is used as a signal generator, and DAB as the chromogen, to stain PCNA-containing nuclei a dark brown. This kit also contains 5 PCNA positive control slides for the convenience of the investigator; 1 stained reference slide and 4 unstained slides for use in the procedure. STORAGE AND SHELF LIFE Store kit at 2-8°C. All performance claims are void after kit expiration date. PCNA Staining Kit Method REAGENTS SUPPLIED Reagent 1. One dropper bottle (6 mL) of ready-to-use blocking solution Reagent 2. One dropper bottle (6 mL) of ready-to-use biotinylated Ms x PCNA primary antibody Reagent 3. One dropper bottle (6 mL) of ready-to-use streptavidin-peroxidase Reagent 4A. One dropper bottle (2 mL) of concentrated substrate buffer (20x) Reagent 4B. One dropper bottle (2 mL) of concentrated chromogen solution, DAB (20x) Reagent 4C. One dropper bottle (2 mL) of 0.6% hydrogen peroxide (20x) Reagent 5. One dropper bottle (6 mL) of ready-to-use Hematoxylin Reagent 6. One dropper bottle (6 mL) of ready-to-use Histomount™ Also contains: 5 Control Slides – 1 stained, 4 unstained REAGENTS AND MATERIALS REQUIRED BUT NOT PROVIDED – Phosphate Buffered Saline (PBS) – Alcohol and xylene – Distilled or deionized water – Quenching solution for endogenous peroxidase – Coverslips SUGGESTED STAINING PROTOCOL A. PRELIMINARY PREPARATION For Paraffin-Embedded Tissues 1. Fix target tissue in 10% NBF (Neutral Buffered Formalin), or in an alcohol-based fixative (such as alcohol or Methacarn). Process tissues for paraffin embedding. 2. Cut tissue 3-4 microns and place on Invitrogen’s HistoGrip™ (Cat. No. 00-8050) or poly-L-lysine coated slides. Dry slides in a 60°C oven for 1-2 hours. 3. Deparaffinize slides in 2 changes of xylene for 5 minutes each. Rehydrate slides in a series of graded alcohol. 4. (Optional): Block for endogenous peroxidase activity with 3% H2O2 in methanol for 10 minutes. Rinse slides in 3 changes of PBS for 2 minutes each. 5. Slides are now ready for PCNA staining. 6. (Optional): Specific staining of formalin-fixed, paraffin-embedded tissues may be enhanced using a Heat Induced Epitope Retrieval (HIER) method. 7. (Optional): If alcohol-fixed tissues are used, tissue sections can be post-dipped in 10% NBF for 30-60 seconds. Rinse well. Fix cells in 70% alcohol or acid-ethanol for 15-20 minutes at 4°C. Acetone or Methacarn fixatives can also be used. 2. If necessary, block for endogenous peroxidase activity with 3% H2O2 in methanol for 10 minutes. 3. Wash in 3 changes of PBS for 2 minutes each. SUGGESTED STAINING PROTOCOL (CONTINUED) B. STAINING PROTOCOL Reagent Specimen Preparation 1. BLOCKING SOLUTION Ready-to-use. Reagent 1 2. BIOTINYLATED MOUSE ANTI-PCNA PRIMARY ANTIBODY Ready-to-use. Reagent 2 3. STREPTAVIDIN-PEROXIDSE Ready-to-use. Reagent 3 4. DAB CHROMOGEN Reagents 4A, 4B and 4C 20x concentrated components. 5. HEMATOXYLIN Ready-to-use. Reagent 5 a. Apply 2 drops (100 μL) or enough to completely cover tissue, of BLOCKING SOLUTION to each specimen. b. Incubate at room temperature for 10 minutes. c. Drain or blot off the solution. DO NOT RINSE a. Apply 2 drops (100 μL) or enough to completely cover tissue, of PRIMARY ANTIBODY to each specimen. b. Incubate in moist chamber for 30-60 min. c. Rinse with PBS for 2 min., 3 times. a. Apply 2 drops (100 μL) or enough to completely cover tissue, of STREPTAVIDINPEROXIDASE to each specimen. b. Incubate at room temperature for 10 minutes. c. Rinse with PBS for 2 min., 3 times. a. Add 1 drop of Reagent 4A, 1 drop of Reagent 4B and 1 drop of Reagent 4C to 1 mL distilled or deionized water. Mix well. Protect from light and use within one hour. b. Apply 2 drops (100 μL) or enough to completely cover tissue, of DAB CHROMOGEN to each section. Incubate for 2-5 min. a. Apply 2 drops (100 μL) or enough to completely cover tissue, of HEMATOXYLIN to each specimen. Incubate for 1-2 min. b. Wash slides in tap water. c. Put slides into PBS until sections turn blue (approx. 30 seconds) d. Rinse in distilled water a. Dehydrate slides in a graded series of alcohol, and clear with xylene. b. Reagent 6 TROUBLESHOOTING Possible causes for negative staining on positive slides: 1. Steps in the staining protocol were performed in incorrect sequence. 2. Primary or secondary antibody incubation steps were omitted. 3. Labile antigens were destroyed. 4. Specimen was improperly fixed and/or processed. 5. Specimen dehydrated during staining. Possible causes for weak staining on all slides: 1. Specimen retained excess liquid after rinsing steps. 2. Incubation times were insufficient. 3. Substrate prepared improperly. 4. Deparaffinization was incomplete (staining may be accompanied by high background). Possible causes for high background staining: 1. Endogenous peroxidase activity was incompletely blocked. 2. Deparaffinization was incomplete. 3. Excessive application of tissue adhesive. 4. Inadequate rinsing of slides. 5. Over-development of substrate. 6. Dehydration of specimen during staining. Los paquetes de tinción de PCNA se utilizan para la detección cualitativa de la proteína PCNA en tejidos preparados en parafina y fijados con formalina y en tejidos congelados. ALMACENAMIENTO Y CONSERVACIÓN Guardar el paquete entre 2°C a 8°C. Todas las reclamaciones sobre rendimiento serán nulas pasada la fecha de caducidad. REACTIVOS SUMINISTRADOS Reactivo 1. PREPARACIÓN PRELIMINAR Tejidos preparados en parafina 1. Fijar el tejido tratado en NBF (formalina neutra tamponada), o en un fijador de alcohol (como alcohol o Metharcarn). Proceder con la inclusión en parafina. 2. Cortar el tejido con un grosor de 3 a 4 micras y colocarlo en HistoGrip™ de Invitrogen (nº. de cat. 00-8050) o cubreobjetos cubiertos con poliL-lisina. Secar los portaobjectos en un horno a 60ºC de 1 a 2 horas. 3. Proceder con la desparafinación de los cubreobjetos en dos baños de xileno durante 5 minutos cada uno. Rehidratar los portaobjetos en una serie de alcohol gradual. 4. (Opcional): Bloquear la actividad de la peroxidasa endógena con H2O2 (peróxido de hidrógeno) al 3% en metanol durante 10 minutos. Enjuagar los cubreobjetos en 3 baños de PBS durante 2 minutos cada uno. 5. Los portaobjetos están preparados ahora para la tinción de PCNA. 6. (Opcional): La tinción específica de tejidos fijados con formalina y con inclusión en parafina pueden potenciarse utilizando un método de Eliminación de Epítope Inducida por Calor. 7. (Opcional): Si se utilizan tejidos fijados con alcohol, los cortes de tejidos pueden pasar por un baño posterior en NBF al 10% de 30 a 60 segundos. Enjuagar bien. Esto puede ayudar a mejorar la morfología. Para la células cultivadas y suspensiones celulares 1. Fijar las células en alcohol al 70% o en ácido-etanol de 15 a 20 minutos a 4ºC. También se pueden usar fijadores de acetona o Methacarn. 2. En caso necesario, bloquear la actividad de la peroxidasa endógena con H2O2 (peróxido de hidrógeno) al 3% en metanol durante 10 minutos. 3. Añadir 2 gotas (100 μL) de SOLUCIÓN DE BLOQUEO a cada muestra, o lo suficiente como para cubrir completamente el tejido. b. Incubar a temperatura ambiente durante 10 minutos. c. Drene o seque suavemente la solución con papel absorbente. NO ENJUAGAR. a. Añadir 2 gotas (100 μL) de ANTICUERPO PRIMARIO a cada muestra, o lo suficiente como para cubrir completamente el tejido. b. Incubar en una cámara de humedad durante 30-60 minutos. c. Enjuagar con PBS (2 minutos, 3 veces). a. Añadir 2 gotas (100 μL) de STREPTAVIDINA-PEROXIDASA a cada muestra, o lo suficiente como para cubrir completamente el tejido. b. Incubar a temperatura ambiente durante 10 minutos. c. Enjuagar con PBS (2 minutos, 3 veces). a. Añadir 1 gota del reactivo 4A, 1 gota del reactivo 4B y 1 gota del reactivo 4C a 1 mL de agua destilada o desionizada. Mezclar bien. Proteger de la luz y usar durante la hora siguiente. b. Añadir 2 gotas (100 μL) de CROMÓGENO DAB a cada corte, o lo suficiente como para cubrir completamente el tejido. Incubar durante 2-5 minutos. a. Añadir 2 gotas (100 μL) de HEMATOXILINA a cada muestra, o lo suficiente como para cubrir completamente el tejido. Incubar durante 1-2 minutos. b. Lave los portaobjetos en agua corriente. c. Ponga los portaobjetos en PBS hasta que en los cortes aparezca el azul (aproximadamente 30 segundos). d. Enjuagar en agua destilada. a. Rehidratar los portaobjetos en una serie de alcohol gradual, o limpiar con xileno. b. Añadir 2 gotas (100μL) de HISTOMOUNT y colocar un cubreobjetos. 2. Reactivo 6 TIEMPO DE INCUBACIÓN (minutos) 10 30-60 10 2-5 1-2 DETECCIÓN Y DIAGNÓSTICO DE AVERÍAS Causas posibles de la tinción negativa en portaobjetos positivos: 1. Los pasos en el protocolo de tinción se realizaron en una secuencia incorrecta. 2. Se omitieron los pasos de incubación del anticuerpo primario o secundario. 3. Se destruyeron antígenos lábiles. 4. La muestra no se fijó y/o procesó correctamente. 5. La muestra se deshidrató durante la tinción. Causas posibles de la tinción débil en todos los portaobjetos: 1. La muestra retuvo exceso de líquido después de los aclarados. 2. Los tiempos de incubación no fueron suficientes. 3. El sustrato no se preparó correctamente. 4. La desparafinización fue incompleta (la tinción puede acompañarse por un fondo elevado). Causas posibles para la tinción elevada del fondo: 1. La actividad de la peroxidasa endógena no se bloqueó completamente. 2. La desparafinización fue incompleta. 3. Aplicación excesiva del adhesivo tisular. 4. Aclarado inadecuado de los portaobjetos. 5. Les équipements de coloration PCNA sont utilisés pour la détection qualitative de la protéine PCNA de tissus de formaline-fixée, paraffine-incluse et gelés. Contient aussi : 5 Lames de Contrôle – 1 colorant, 4 décolorants REACTIFS ET MATERIELS NECESSAIRES MAIS PAS FOURNIS – Phosphate Tamponné Salin (PBS) – Alcool et xylène – Eau distillée ou déminéralisée – Solution refroidissant pour peroxydase endogène – Couvertures PROTOCOLE DE COLORATION SUGGERE A. PREPARATION PRELIMINAIRE Pour des Tissus de Paraffine-Incluse 1. Fixer le tissu cible dans 10% devNBF (Formaline Neutre Tamponnée), ou dans un alcool à base fixative (comme l'alcool ou le Méthacarn). Préparer les tissus pour la paraffine incluse. 2. Couper le tissu de 3-4 microns et placer sur Invitrogen’s HistoGrip™ (Cat. No. 00-8050) ou lames enduites de poly-L-lysine. Sécher les lames dans un four à 60°C pendant 1-2 heures. 3. Déparaffiner les lames 2 fois dans du xylène pendant 5 minutes à chaque fois. Réhydrater les lames dans une série graduée d'alcool. 4. (Optionnel): Bloquer l'activité de peroxydase endogène avec 3% H2O2 dans du méthanol pendant 10 minutes. Rincer les lames dans du PBS pendant 2 min., 3 fois. 5. Les lames sont maintenant prêtes pour une coloration PCNA. 6. (Optionnel): La coloration spécifique de formaline-fixée, tissus de paraffine-incluse peut être potentialisée en utilisant une méthode de Recherche d'Epitope de Chaleur par Induction (HIER). 7. (Optionnel): Si des tissus d'alcool-fixé sont utilisés, des sections de tissu peuvent être trempées dans 10% de NBF pendant 30-60 secondes. Bien rincer. Ceci peut améliorer la morphologie. Pour Cellules de Culture et Cellules en Suspension 1. Fixer les cellules dans 70% d'alcool ou d'acide-éthanol pendant 15-20 minutes à 4°C. L'Acétone ou le Méthacarn fixatifs peuvent être utilisés. 2. Si nécessaire, bloquer l'activité endogène de peroxydase avec 3% H2O2 dans du méthanol pendant 10 minutes. 3. PROTOCOLE DE COLORATION Réactif Préparation de Spécimens : 1. SOLUTION BLOQUANTE Prêt à l'utilisation. Réactif 1 a. Appliquer 2 gouttes (100 μL) ou suffisamment pour couvrir complètement le tissu de SOLUTION BLOQUANTE à chaque spécimen. b. Incuber les lames à température ambiante pendant10 minutes. c. Egoutter ou sécher la solution. NE PAS RINCER a. Ajouter 2 gouttes (100 μL) ou suffisamment pour couvrir complètement le tissu de chaque spécimen d'ANTICORPS PRIMAIRE. b. Incuber dans une chambre humide pendant 30-60 min. c. Rincer avec du PBS pendant 2 min., 3 fois. a. Appliquer 2 gouttes (100 μL) ou suffisamment pour couvrir complètement le tissu de chaque section de STREPTAVIDINE-PEROXYDASE. b. Incuber les lames à température ambiante pour 10 minutes. c. Rincer avec du PBS pendant 2 min., 3 fois. a. Ajouter 1 goutte de Réactif 4A, 1 goutte de Réactif 4B et 1 goutte de Réactif 4C à 1 mL d'eau distillée ou déminéralisée. Bien mélanger. Protéger de la lumière et à utiliser dans l'heure. b. Appliquer 2 gouttes (100 μL) ou suffisamment pour couvrir complètement le tissu de CHROMOGENE DAB de chaque section. Incuber pendant 2-5 min. a. Appliquer 2 gouttes (100 μL) ou suffisamment pour couvrir complètement le tissu de chaque spécimen d'HEMATOXYLINE. Incuber pendant 1-2 min. b. Laver les lames sous l'eau du robinet. c. Mettre les lames dans du PBS jusqu'à ce qu'elles deviennent bleues (approx. 30 secondes) d. Rincer dans de l'eau distillée a. Déshydrater les lames en série graduée d'alcool et clarifier dans du xylène. b. Ajouter 2 gouttes (100 μL) d'HISTOMOUNT et couvrir. 2. Les étapes du protocole de coloration n’ont pas été exécutées dans le bon ordre. 2. Les étapes d’incubation des anticorps primaires ou secondaires ont été omises. 3. Les antigènes labiles ont été détruites. 4. Le spécimen était mal attaché et/ou traité. 5. Le spécimen s’est désydraté pendant la coloration. Causes possibles pour une coloration faible sur toutes les lames : 1. Le spécimen a retenu un excès de liquide après les étapes de rinçage. 2. Les temps d’incubation n’étaient pas suffisants. 3. Le substrat était mal préparé. 4. La déparaffination était incomplète (la coloration peut être accompagnée d’une coloration de fond élevée). Causes possibles pour une coloration de fond élevée : 1. L’activité de la peroxydase endogène n’était pas complètement bloquée. 2. La déparaffination était incomplète. 3. Application excessive de l’adhésif tissulaire. 4. Mauvais rinçage des lames. 5. Surdéveloppement du substrat. 6. Déshydratation du spécimen pendant la coloration. L’interpretazione deve essere effettuata da un patologo qualificato entro il contesto dell’anamnesi clinica del paziente ed in considerazione di altri test diagnostici. IIkit di colorazione PCNA sono utilizzati per la rilevazione qualitativa della proteina PCNA in tessuti fissati in formalina, inclusi in paraffina e tessuti congelati. PREPARAZIONE PRELIMINARE Per tessuti inclusi in paraffina 1. Fissare il tessuto bersaglio in 10% NBF (Formalina neutra tamponata), o in un fissante a base di alcool (come alcool oppure Methacarn). Preparare i tessuti per l’inclusione in paraffina. 2. Tagliare i tessuti 3-4 micron e porli su Invitrogen’s HistoGrip™ (Cat. n.. 00-8050) oppure vetrini rivestiti di poli-L-lisina. Far asciugare i vetrini in forno a 60 °C per 1-2 ore. 3. Sparaffinizzare i vetrini in 2 cambi di xilene per 5 minuti ciascuno. Reidratare i vetrini in una scala crescente di alcool. 4. (Facoltativo): Bloccare per perossidasi endogena con 3% H2O2 in metanolo per 10 minuti. Risciacquare i vetrini con 3 cambi di PBS per 2 minuti ciascuno. 5. I vetrini sono ora pronti per la colorazione PCNA. 6. (Facoltativo): colorazione specifica di tessuti fissati in formalina, inclusi in paraffina può essere evidenziata utilizzando un metodo Recupero di epitopi mediante calore (Heat Induced Epitope Retrieval) (HIER). 7. (Facoltativo): se si utilizzano tessuti fissati in alcool, le sezioni di tessuto si possono immergere poi in 10% di NBF per 30-60 secondi. Risciacquare bene. Questo potrebbe migliorare la morfologia. Per Cellule Coltivate e Sospensioni cellulari 1. Fissare le cellule in 70% di alcool oppure acido-etanolo per 15-20 minuti a 4 °C. Si possono utilizzare acetone oppure fissativi Methacarn. 2. Se necessario, bloccare per la perossidasi endogena con 3% H2O2 in metanolo per 10 minuti. 3. PROTOCOLLO DI COLORAZIONE Reagente Preparazione dei campioni 1. SOLUZIONE BLOCCANTE Pronto all’uso. Reagente 1 a. Applicare a ciascun campione 2 gocce (100 μL) di SOLUZIONE BLOCCANTE oppure una quantità sufficiente a ricoprire completamente il tessuto. b. Incubare a temperatura ambiente per 10 minuti. c. Rimuovere con tampone la soluzione in eccesso. NON RISCIACQUARE a. Applicare a ciascun campione 2 gocce (100 μL) di ANTICORPO PRIMARIO oppure una quantità sufficiente a ricoprire completamente il tessuto. b. Incubare in camera umida per 30-60 min. c. Risciacquare con PBS per 2 min., 3 volte. a. Applicare a ciascun campione 2 gocce (100 μL) di STREPTAVIDINA-PEROSSIDASI oppure una quantità sufficiente a ricoprire completamente il tessuto. b. Incubare a temperatura ambiente per 10 minuti. c. Risciacquare con PBS per 2 min, 3 volte. a. Aggiungere 1 goccia di Reagente 4A, 1 goccia di Reagente 4B e 1 goccia di Reagente 4C ad 1 mL di acqua distillata oppure deionizzata. Mescolare bene. Conservare al riparo della luce e utilizzare entro un’ora. b. Applicare a ciascuna sezione 2 gocce (100 μL) di CROMOGENO DAB oppure una quantità sufficiente a ricoprire completamente il tessuto. Incubare per 2-5 min. a. Applicare a ciascun campione 2 gocce (100 μL) di EMATOSSILINA oppure una quantità sufficiente a ricoprire completamente il tessuto. Incubare per 1-2 min. b. Lavare i vetrini con acqua di fonte. c. Mettere i vetrini in PBS sino a che le sezioni diventano blu (circa 30 secondi) d. Risciacquare in acqua distillata. a. Deidratare i vetrini in una scala crescente di alcool, e chiarificare con xilene. b. Aggiungere 2 gocce (100 μL) di HISTOMOUNT e mettere il coverslip. 2. Le fasi previste dal protocollo di colorazione non sono state eseguite nell’ordine corretto. 2. Sono state omesse le fasi di incubazione dell’anticorpo primario e secondario. 3. Sono stati distrutti degli anticorpi labili. 4. I provini non sono stati debitamente fissati e/o trattati. 5. I provini si sono disidratati nel corso della colorazione. Cause possibili di colorazione debole in tutti i vetrini: 1. I provini hanno trattenuto il liquido in eccesso accumolatosi durante le fasi di risciacquo. 2. Sono stati osservati tempi di incubazione insufficienti. 3. Il substrato non è stato preparato correttamente. 4. Deparaffinizzazione incompleta (la colorazione potrebbe essere accompagnata da fondo elevato). Cause possibili di colorazione con fondo elevato: 1. L’attività perossidasica endogena non è stata bloccata completamente. 2. Deparaffinizzazione incompleta. 3. Applicazione di un quantitativo eccessivo di adesivo per tessuti. 4. Risciacquo inadeguato dei vetrini. 5. Sovrasviluppo del substrato. 6. Disidratazione dei provini durante la colorazione. VORLÄUFIGE PRÄPARATION Für Paraffin-eingebettete Gewebe 1. Zielgewebe in 10%gem NBF (Neutral gepuffertes Formalin), oder in einem Alkohol-basierten Fixiermittel (wie Alkohol oder Methacarn) fixieren. Gewebe für Paraffineinbettung verarbeiten . 2. Gewebe 3-4 Microne schneiden und sie auf Invitrogen’s HistoGrip™ (Cat. Nr. 00-8050) oder Poly-L-lysine beschichteten Objektträgern plazieren . Objektträger in einem 60°C Ofen für 1-2 Stunden trocknen. 3. Objektträger in 2maligem Wechsel von Xylene jeweils für 5 Minuten deparaffinisieren. Objektträger in einer Reihe von abgestuftem Alkohol rehydrieren. 4. (Optional): Blockieren Sie für endogene Peroxidasen Aktivität mit 3% H2O2 in Methanol für 10 Minuten. Spülen Sie Objektträger in 3maligen Wechseln von PBS für jeweils 2 Minuten. 5. Objektträger sind jetzt fertig für PCNA Färbung. 6. (Optional): Spezifisches Färben von Formalin-fixierten, Paraffin-eingebetteten Geweben kann durch die Benutzung einer Hitze induzierten Epitop Wiedergewinnungsmethode (HIER) verstärkt werden. 7. (Optional): Wenn Alkohol-fixierte Gewebe benutzt werden, können Gewebesektionen in 10%igem NBF für 30-60 Sekunden hinterher eingetaucht werden. Gut spülen. Dieses kann die Morphologie verbessern. Für Kultivierte Zellen und Zell Suspensionen 1. Zellen in 70%igem Alkohol oder Acid-Aethanol für 15-20 Minuten 4°C fixieren. Azetone oder Methacarn Fixiermittel können ebenfalls benutzt werden. 2. Vor Licht schützen und innerhalb einer Stunde benutzen. b. 2 Tropfen (100 μL) von DAB CHROMOGEN auf jede Sektion geben oder genug, um das Gewebe vollständig zu bedecken. Für 2-5 Min. inkubieren. a. 2 Tropfen (100 μL) von HAEMATOXILIN auf jede Probe geben oder genug, um das 1-2 Gewebe vollständig zu bedecken. Für 1-2 Min. inkubieren. b. Die Objektträger in Leitungswasser waschen. c. Die Objektträger in PBS legen bis sie blau werden (ungefähr. 30 Sekunden) d. Gut in destilliertem Wasser spülen. a. Reagens 6 FEHLERBEHEBUNG Mögliche Ursachen für eine negative Färbung auf positiven Objektträgern: 1. Die Reihenfolge der Schritte des Färbungsprotokolls wurde nicht eingehalten. 2. Die Inkubationsschritte für den primären oder sekundären Antikörper wurden ausgelassen. 3. Labile Antigene wurden zerstört. 4. Die Probe wurde nicht richtig fixiert und/oder verarbeitet. 5. Die Probe trocknete während der Färbung aus. Mögliche Ursachen für eine schwache Färbung auf allen Objektträgern: 1. Nach der Spülung blieb zuviel Flüssigkeit in der Probe zurück. 2. Die Inkubationszeiten waren nicht ausreichend. 3. Das Substrat wurde falsch vorbereitet. 4. Unvollständige Deparaffinierung (Färbung ist von einer starken Hintergrundfärbung begleitet). Mögliche Ursachen für starke Hintergrundfärbung: 1. Endogene Peroxidase-Aktivität wurde unvollständig blockiert. 2. Unvollständige Deparaffinierung. 3. Übermäßiger Gebrauch von Gewebefixiermittel. 4. Unzureichende Spülung der Objektträger. 5. Überentwicklung des Substrats. 6. Dehydrierung der Probe während der Färbung.
