NOTE: Taq Polymerase obtained from a licensed manufacturer should be used when performing this assay. This kit is based on the co-amplification of a 147bp product from the Factor V gene, a 266bp product from the Factor II gene and an internal control product of 650bp. STORAGE AND SHELF-LIFE Store the kit at -20°C. Prior to use thaw reagents at 15...30°C. Mix each stock thoroughly and whenever possible keep on ice during use. Refreeze unused reagents. NOTE: Do not warm in a waterbath. 350bp (HIC) 300bp (HIC) STEP-BY-STEP PROCEDURE PCR Method 1. Add 2µl of Taq polymerase (5 units/µl) to 250µl FII&V MasterMix (tube 1) = Blue MasterMix (BMM). Mix by pipetting. BMM can now used in step 2 or it can be stored at -20°C for up to 12 months. 2. Pipette 23µl of BMM into reaction tube. Add 1µl of HIC. Add 1µl of control DNA or genomic DNA (50-100 ng). Mix by pipetting. NOTE: Dilute DNA with 10/1 TE if necessary. 3. Overlay with mineral oil if using a thermal cycler without a heated lid. 4. Place tubes in the thermal cycler. Cycle as follows: Initial denaturation at 95°C for 3 mins followed by 30 cycles of: 95°C for 30 secs 63°C for 60 secs 72°C for 60 secs Restriction Digestion 5. Add 1µl of Hind III/Mnl I mixture. Mix by pipetting and incubate at 37°C for 3 hrs. Load 10µl of the digested sample onto 1.7% agarose gel (containing ethidium bromide) and electrophorese next to 10µl of 100 bp DNA ladder. There is no need to add loading buffer. REMARQUE : Utiliser de la Taq polymérase provenant d’un fabricant autorisé lorsque vous réalisez cette analyse. Autres composants du kit Chaque kit contient également 1 fiche technique. STOCKAGE ET CONSERVATION Conserver le kit à -20°C. Décongeler les réactifs entre 15...30°C avant utilisation. Bien mélanger chaque produit et maintenir, si possible, sur la de la glace pendant l’utilisation. Recongeler les réactifs non utilisés. REMARQUE : Ne pas chauffer dans un bain-marie. La figure 1, ci-dessous, représente les résultats du protocole de détection de mutation multiple du facteur V de Leiden et du facteur II (G>A 20210) sur un gel d’agarose HB-ADN à 1,7% ; de gauche à droite. 350bp (CIH) 300bp (CIH) MÉTHODOLOGIE Méthode PCR 1. Ajouter 2 µl de Taq polymérase (5 unités/µl) à 250 µl de MasterMix F II et F V (tube 1) = MasterMix bleu (MMB). Mélanger à la pipette. Il est alors possible d’utiliser le MMB pour l’étape 2 ou de le conserver à -20°C pendant 12 mois. 2. Pipeter 23 µl de MMB dans un tube à essai. Ajouter 1 µl de CIH. Ajouter 1 µl d’ADN de contrôle ou d’ADN génomique (50 – 100ng). Mélanger à la pipette. REMARQUE : Diluer l’ADN avec du 10/1 TE si nécessaire. 3. Couvrir d’une couche d’huile minérale si vous utilisez un thermocycleur sans couvercle chauffant. 4. Placer les tubes dans le thermocycleur. Programmer de la façon suivante : dénaturation initiale à 95°C pendant 3 minutes, suivie de 30 cycles de : 30 secondes à 95°C 60 secondes à 63°C 60 secondes à 72°C Digestion par des enzymes de restriction 5. Ajouter 1µl de mélange Hind III/Mnl I. Mélanger à la pipette et incuber à 37°C pendant 3 heures. Électrophorèse 6. Charger 10µl d’échantillon digéré sur un gel d’agarose à 1,7% (contenant du bromure d’éthidium) et faire migrer à côté de 10µl d’échelle d’ADN 100pb. Il n’est pas nécessaire d’ajouter un tampon de charge. Afin de séparer les fragments de 266 et de 238pb, il est nécessaire de traiter les échantillons jusqu’à ce que le colorant bleu ait migré d’au moins 6cm. 266bp (FII) 238bp (FII) 122bp (FV) Piste 1. Elektrophorese 6. 10 µl der mit Restriktionsenzym behandelten Probe auf ein 1,7% Agarose-Gel (mit Ethidiumbromid) neben 10 µl eines 100 bp DNA Längenstandards auftragen und Elektrophorese durchführen. Zufügen eines Auftragspuffers ist nicht erforderlich. Zum Trennen der 266 und 238 bp Fragmente müssen die Proben solange laufen, bis die blaue Farbe mindestens 6 cm in das Gel diffundiert ist. Fila 1: Fila 2: 350bp (HIC) PROCEDURA Metodo PCR 1. Aggiungere 2µl di Taq polimerasi (5 unità/µl) a 250µl di FII&V MasterMix (provetta 1) = Blue MasterMix (BMM). Miscelare pipettando. Ora il BMM può essere utilizzato nella fase 2 oppure può essere conservato a -20°C fino a 12 mesi. 2. Pipettare 23µl di BMM nella provetta di reazione. Aggiungere 1µl di HIC.
