The Thermo Scientific Dewax and HIER Buffer L is intended for use in immunohistochemistry. AVAILABILITY Catalog # TA-999-DHBL TA-100-DHBL Volume 1000ml (15x) 100ml (15x) Quantity 10X100ml 1X100ml DESCRIPTION The Dewax and HIER Buffer L is designed to simultaneously deparaffinize/dewax and perform heat induced epitope retrieval (HIER) on formalin-fixed paraffin-embedded (FFPE) tissue sections. FFPE tissue sections can be processed through the Dewax and HIER Buffer L, prior to manual staining or automated staining on the Thermo Scientific Autostainer. Formaldehyde fixation negatively impacts immunoreactivity of antigens/epitopes. While enzymatic digestion reverses this status for only a few antigens/epitopes, HIER has been established as the pre-treatment method of choice for many antigens/epitopes to improve their immunoreactivity with antibodies designed to detect their expression in FFPE tissues. After dilution, the Dewax and HIER Buffer L can be used for a maximum of three runs within five days when mounting the slides onto the Autostainer slide racks and processing them in the Thermo Scientific PT Module. Please refer to the instruction for use of the individual antibody for the recommended pretreatment method. WARNINGS & PRECAUTIONS Refer to MSDS. STORAGE & SHELF LIFE When stored at room temperature the product is stable for 1 year. Do not use after the indicated expiration date. After dilution, the buffer can be used for a maximum of three runs within five days when mounting the slides onto the Autostainer slide racks and processing them in the PT Module. PROCEDURE This is a 15x concentrate solution and must be diluted 1:15 with deionized water before use. use solution should be 6.2 ± 0.2. 1. 2. Pour one bottle of Dewax and HIER Buffer L (100ml) into a PT Module tank; add 1,400ml of deionized water to obtain 1,500ml (1.5 liters) of ready-to-use solution (1:15 dilution). Measure and document pH with calibrated pH meter per accreditation guidelines (optional). For antigen retrieval, pre-heat the PT Module to 85°C and immerse the slides mounted on the Autostainer slide rack into the PT Module tank. Start the PT Module program; it will warm the buffer and the slides to 98°C and hold this temperature for 20 minutes. Cool slides in the PT Module to 85°C. Cool further to room temperature in TBST buffer for at least 5 minutes. Ensure slides are kept moist in buffer until loaded onto the Autostainer. Use Thermo Scientific Richard-Allan Scientific Parapel Paraffin Repellent (Cat # 2300) to keep PT Module tank clean of wax residue. Il tampone L per sparaffinatura e smascheramento antigenico Thermo Scientific è ideato per l’utilizzo in immunoistochimica. DISPONIBILITA’ CODICE TA-999-DHBL TA-100-DHBL VOLUME 1000ml (15x) 100ml QUANTITA’ 10x100ml 1x100ml DESCRIZIONE Il tampone L per sparaffinatura e smascheramento antigenico Thermo Scientific è ideato per sparaffinare e realizzare contemporaneamente lo smascheramento antigenico indotto da calore (HIER) in sezioni di tessuti fissati in formalina ed inclusi in paraffina. Le sezioni possono essere trattate tramite l’uso del tampone L prima della colorazione manuale o automatica nel coloratore automatico Autostainer Thermo Scientific. La fissazione in forlmaldeide influisce negativamente sull’immunoreattività degli antigeni/epitopi. Mentre la digestion enzimatica è in grado di ripristinare questo stato solo per pochi antigeni/epitopi, il tampone HIER è stato designato come il metodo di pre-trattamento di prima scelta per molti antigeni/epitopi per migliorare la loro immunoreattività con anticorpi ideati per rilevare la loro espressione nei tessuti fissati in formalina ed inclusi in paraffina. Dopo la diluizione, il tampone L per sparaffinatura e smascheramento antigenico può essere utilizzato per un massimo di tre trattamenti entro cinque giorni dall’inserimento dei vetrini nel rack dell’Autostainer ed il trattamento nel PT Module Thermo Scientific. Fare riferimento alle istruzioni d’uso del singolo anticorpo per il metodo di pretrattamento raccomandato. AVVERTIMENTI E PRECAUZIONI Fare riferimento alla scheda di sicurezza CONSERVAZIONE E VALIDITA’ Quando conservato a temperature ambiente il prodotto è stabile per un anno. Non usare dopo la data di scadenza indicate. Dopo la diluizione il tampone può essere usato per un Massimo di tre trattamenti entro cinque giorni dall’inserimento dei vetrini nel rack dell’Autostainer ed il trattamento nel PT Module Thermo Scientific. FORNITO COME Soluzione concentrata 15x. PROCEDURA Questa è una soluzione concentrata 15x e deve essere diluita 1:15 con acqua deionizzata prima dell’uso. Il pH della soluzione finale pronta all’uso dovrebbe essere 6.2 ± 0.2. 1. Sezioni fissate in formalina, incluse in paraffina (FFPE) ed asciutte come di norma. 2. Versare una bottiglia di tampone (Dewax and HIER Buffer L) (100ml) in una vasca del PT Module; aggiungere 1400ml di acqua deionizzata per ottenere 1500ml (1,5 litri) di soluzione pronta all’uso (diluizione 1:15). 3. Misurare e registrare il pH con un pH-metro tarato secondo linee guida accreditate (opzionale). 4. Per lo smascheramento antigenico, preriscaldare il PT Module a 85°C ed immergere I vetrini montati sul rack dell’Autostainer nella vasca del PT Module. 5. Avviare il programma del PT Module; scalderà il tampone ed I vetrini a 98°C e manterrà la temperatura per 20 minuti 6. Raffreddare I vetrini nel PT Module a 85°C. Raffreddare ulteriormente fino a temperatura ambiente nel tampone TBST per almeno 5 minuti. 7. Assicurarsi che I vetrini restino bagnati del tampone fino al caricamento nell’Autostainer. 8. Usare il repellente per paraffina Richard-Allan Scientific Parapel Thermo Scientific (codice 2300) per mantenere il PT Module pulito dai residui di paraffina.
