Mechanism of action:
- Tyrosinase plays key role in melanogenesis as it regulates the rate-limiting step of melanogenesis
- 4-n-butylresorcinol has an inhibitory effect on the melanogenesis of cultured B16 melanoma cells by
its direct inhibition of tyrosinase activity, as well as the suppression of tyrosinase synthesis, without
inducing any cytotoxicity.
- Several in vitro studies have demonstrated that 4-n-butylresorcinol inhibits melanin production as well
as the activity of both tyrosinase and tyrosinaserelated protein-1 (TRP-1)

- 1 gm of 4-n-Butylresorcinol is soluble in 3gm of 3 polyglyceryl diisostearate.
Oil in water emulsion is the suitable type of emulsion for creams with 4 butyl resorcinol.
In this type of skin cream formulation the active (4-n-Butylresorcinol) is dissolved in a solvent (e.g.
glycerin) then this needs to be added below 45°C in the formulation post emulsification.
Toxicology data
Acute Oral LD50:
The oral LD50 of butyl resorcinol in the rat, rabbit, guinea pig and dog ranged from 550 to >5000 mg/kg
body weight.
Sub acute toxicity:
Oral doses up to 500 mg/kg to mice and rats showed reduced body weight only for male rats. No other
relevant compound- related findings were noted.
Allergenicity/skin irritation/skin sensitization:
Tests in guinea pigs showed that hexylresorcinol was non-sensitizing when applied topically. HRIPT
(1%, 2% & 5% in corn oil, total subjects 240) demonstrates it to be a non-primary irritant and a nonprimary sensitizer. Skin-irritating propensities using 0.1% and 0.5% level on scarified skin: Showed low
irritating potential and compares well with saline control.
Application
- 4-n-Butylresorcinol is safe to use because it is fragrance free and free from color additives.
- 4-n-Butylresorcinol penetrates quickly and reaching deep into the skin.
- 4-n-Butylresorcinol prevents formation of melanin and more effective than other conventional
lightening products.

Example 1: A depigmenting cream for the skin of oil-in-water emulsion type is prepared, which cream
comprises (% by weight)
Content: N butyl resorcinol 3% Glycerol stearate 2% Polysorbate 60 (Tween 60 from ICI) 1% Stearic
acid 1.4%.sup. Triethanolamine 0.7%.sup.Carbomer 0.4%.sup. Hydrogenated polyisobutene 12%
Perhydrosqualene 12% Glycerol 5% Antioxidant 0.05% Preservative q.s. Water q.s. for 100%
Example 2: A depigmenting gel for the skin is prepared, which gel comprises (% by weight)
Content: N Butyl resorcinol 5% Hydroxypropylcellulose 1% Antioxidant 0.05% Isopropanol 40%
Preservative q.s. Water q.s. for 100%
IMPORTANT SUGGESTION:
The product is very sensitive to light, humidity and oxidation.
When you formulate with n-butylresorcinol it is very imortant to use a powerful anti-oxidant to protect it
(BHT is the best choice, tochopherol could be a suitable alternative, while citric acid is not working).
Is it possible to observe a slight pinkish color of the product, this is normal and it is not compromising
the efficacy; if the color become light brown or brown this means the product is not been stored or
handled in the correct way, it is oxidized and it lost his efficacy.
